The transgenic RNAi in flies is the most convenient tool to study the genes in vivo through conditional manner, and as an ideal animal model platform to screen novel modulators in signaling pathways and diseases, as most fly genes are homologous to human, and the majority human disease genes can be found in fly. To improve the knockdown efficacy and to limit position effect and off targets as well, we continuously developed the 2nd、 3rd and next generation transgenic RNAi techniques. It significantly triggers RNAi not only in soma, but also in germline (Ni et al., 2008, 2011, Nature Methods). Moreover, controling multiple genes together is achievable (Qiao et al., 2018, Nature Communications). TsingHua Fly Center (THFC) is established in 2011, founded as a non-profit organization to provide resource and technical service. Currently we are keeping thousands of transgenic lines in house for life science and biomedical research.
Besides expanding the transgenic RNAi resources, we also developed CRISPR/Cas9 genome editing system for the fly community (Ren et al., 2013, PNAS). We reported an effective CRISPR-based method for genome editing in Drosophila. In this system, plasmids encoding single-guide RNAs (sgRNAs) are injected into embryos of germline specific Cas9 transgenic flies. The major features of our system are its high heritable efficiency and ease to apply.
As to gene overexpression(activation), we developed flySAM(Jia et al.,2018, PNAS), which is a transcriptional activation technology based on CRISPR/Cas9.
